Abstract

SEPALLATA(SEP)-like,a class E MADS-box transcription factor gene,plays a key role in development of flower organs.In order to investigate the function of SEP-like gene in the spur development of Impatiens uliginosa,the genes MADS-EJ2 and MADS2 were successfully cloned in the present study,named IuMADS-EJ2 and IuMADS2,and both of them had a cDNA sequence length of 762 bp and encoded for 253 a-mino acids.Bioinformatic analysis demonstrated that IuMADS-EJ2 was a hydrophilic stable protein,and Iu-MADS2 was a hydrophilic unstable protein,both of them contained MIKC structure and had their sub-family-specific SEPⅠ and SEPⅡ motifs at C terminus,which were class E genes.Subcellular localization predicted that IuMADS-EJ2 and IuMADS2 were located in nuclei.The amino acid sequences of IuMADS-EJ2 and Iu-MADS2 genes were both clustered with the homologous gene of Impatiens glandulifera with the similarity of 98.03%and 95.26%respectively.The qRT-PCR analysis indicated that both the two genes expressed at three development periods and two different parts of I.uliginosa spur.IuMADS-EJ2 expressed the highest in the pet-al spur at the early stage,and IuMADS2 expressed the highest in the limb at the early stage.It was speculated that the two genes regulate the development of petal spur,especially at the early stage of petal spur develop-ment.The results could provide theoretical bases for the development mechanism analysis of petal spur,the improvement of flower form and the new variety cultivation of Impatiens.
