摘要

Abstract

Objective:To investigate the effects of a high-salt diet on arterial blood pressure,left ventricular function,cardiac vagal nerve reflex,myocardial injury and the expression of inflammatory factors in rats,and to clari-fy the comprehensive impact of high-salt diet on cardiac physiological function and its mechanism.Methods:SD rats were randomly divided into a control group and a high-salt group.Rats in the control group were fed a normal diet,while those in the high-salt group were fed high-salt diet for a duration of 8 weeks.Arterial blood pressure of rats was monitored at weeks 0,1,3,5 and 7 using the non-invasive tail artery blood pressure measurement technique.At the end of the 8th week,following anesthesia,left ventricular hemodynamics were assessed via left ventricular intuba-tion through the right common carotid artery.The effects of the cardiac vagus nerve reflex on blood pressure and left ventricular pressure were observed after applying capsaicin to the myocardial outer membrane.Electrocardiograms were recorded using limb leads.Serum myoglobin(MYO)and hypersensitive cardiac troponin(hs-cTn)levels were measured using ELISA,meanwhile the expression of atrial natriuretic peptide(ANP),myosin heavy chain 6(MYH6),myosin heavy chain 7(MYH7)and inflammatory factors in the heart were detected by qRT-PCR.Results:Compared with the control group,the systolic blood pressure(SEP),diastolic blood pressure(DBP)and mean arteri-al pressure(MAP)of rats in the high-salt group showed a continuous increase starting one week after the high-salt diet(all P<0.05).After 8 weeks,the left ventricular end-diastolic pressure(LVEDP)increased significantly,and the maximum rate of increase(dP/dtmax)and the absolute value of maximum rate of decrease(-dP/dtmax)of left ventricular pressure were notably lower(all P<0.01).The change rates of SBP,MAP,LVDEP and left ventricular systolic blood pressure(LVSP)in the high-salt group rats after capsaicin application were significantly lower than those in the control group(all P<0.05).Concurrently,the high-salt group exhibited abnormal electrocardiogram changes,including ST-segment elevation,wide and malformed QRS waves,and atrial premature beats with atrioven-tricular block.Additionally,serum concentrations of MYO and hs-cTn increased significantly in the high-salt group(P<0.05).The mRNA expressions of ANP,MYH7,interleukin-1β(IL-1β),interleukin-6(IL-6)and cyclooxygen-ase 2(COX2)in the heart were upregulated,while that of MYH6 decreased(all P<0.05).Conclusion:High salt in-take contributes to myocardial contractile and diastolic dysfunction in rats,coupled with reduced efferent sensitivity of the cardiac vagus nerve.This mechanism may be attributed to myocardial inflammatory injury.
