摘要

Abstract

In order to explore the optimum extraction process of Periplaneta americana polypeptide and evaluate its immune activity,three different extraction methods of ultrasonic cell disruption,trypsin digestion and neutral protease digestion were compared by using Periplaneta americana as the raw material.It was found that trypsin digestion had the highest extraction efficiency.The effects of enzyme-to-substrate ratio,solid-liquid ratio and ultrasonic time on the yield of Periplaneta americana polypeptide were further investigated.The effects of Periplaneta americana polypeptide on the proliferation and phagocytic activity of RAW264.7 cells were detected by CCK-8 method and neutral red phagocytosis assay.The results showed that the yield of Periplaneta americana polypeptide was(11.92±0.31)%under the conditions of enzyme-substrate ratio 5 000 U/g,solid-liquid ratio 1∶20 g/mL and ultrasonic time 5 h.The results of CCK-8 and neutral red phagocytosis experiments showed that RAW264.7 cells had better proliferation and phagocytic activity when the mass concentration was 5 to 40 μg/mL.The results of in vivo experiments also showed that Periplaneta americana polypeptide showed good immune activity in mice.
