摘要

Abstract

To investigate the effect of MDM2 expression on ferroptosis in colon cancer cells,MDM2 siRNA plasmid and MDM2 overexpression plasmid were transfected into colon cancer cells and the blank control group,empty vector control group,MDM2 interference control group,MDM2 interference group,and MDM2 overexpression group were established.The proliferative activity of cells in each group was detected by CCK8;Fe2+,GSH,MDA content in each group were detected using kit;ROS expression levels in cells from each group were assessed via immunofluorescence;Western blot and RT-qPCR were used to detect the expression levels of MDM2,P53,GPX4,and PTGS2 proteins and mRNAs Tespectively.Compared with the control groups,MDM2 interference significantly increased the levels of MDA,Fe2+,and ROS,as well as the p53 and PTGS2 mRNA/protein expression level,while it significantly decreased the levels of GSH,cell proliferation activity,and the MDM2 and GPX4 mRNA/protein expression.Conversely,MDM2 overexpression significantly decreased the levels of MDA and ROS,as well as the P53 and PTGS2 mRNA/protein expression level,while it significantly increased the levels of GSH,cell proliferation activity,and the MDM2 and GPX4 mRNA/protein expression level.MDM2 inhibits ferroptosis in human colon cancer cells by regulating the p53/GPX4 signaling pathway.
