摘要

Abstract

Objective To investigate the effect and mechanism of eugenol on the growth and virulence of Klebsiella pneumon-iae.Methods K.pneumoniae was inoculated into liquid culture media containing different concentrations(0,125,250 μg/mL)of eugenol.The growth curve and survival rate of the bacteria were measured,and changes in the main virulence fac-tors,including the productions of capsules,pili,and biofilms,were detected.Furthermore,qRT-PCR was performed to determine the mRNA expressions of virulence-related genes.Results After the treatment by eugenol,the growth rate and the survival rate of K.pneumoniae significantly decreased,and these changes were correlated with the dose of eugenol.Ad-ditionally,under the intervention of eugenol,the productions of capsules,pili,and biofilms by K.pneumoniae decreased in a dose-dependent manner.The qRT-PCR indicated that capsule production-related genes(rmpA,magA),biofilm-related gene(bsmA),and pilus synthesis genes(fimA,fimK)were all down-regulated.Moreover,the expression of the global transcription factor(crp)was significantly reduced.Conclusion Eugenol can inhibit the productions of bacterial capsules,pili,and biofilms by acting on CRP.This study provide a theoretical basis and reference for the clinical application of euge-nol as a potential drug to treat K.pneumoniae-induced infectious diseases.
