连翘苷缓解高尿酸血症肾病的作用研究
Effect of Phillyrin on Alleviating Hyperuricemic Nephropathy

Abstract
Objective To investigate the effect of phillyrin on improving hyperuricemic nephropathy.Methods Hyperurice-mia(HUA)mouse model was established by intraperitoneal injection of 300 mg/kg hypoxanthine combined with 300 mg/kg potassium oxonate for 7 days.Forty-two male ICR mice were randomly divided into six groups:a control group,a phill-yrin group(120 mg/kg),a HUA group,a febuxostat group(10 mg/kg),a low-dose phillyrin-HUA group(40 mg/kg),and a high-dose phillyrin-HUA group(120 mg/kg).Serum samples from the mice were tested for uric acid,creatinine,urea nitrogen,and nitric oxide(NO)using a kit method.Gross morphological changes were observed and renal index were calculated.Pathological changes in kidney tissues were analyzed by hematoxylin-eosin(HE)staining.Results The kid-neys in the model group exhibited pathological changes such as significantly increased serum uric acid,tubular dilation,and glomerular atrophy,along with a significantly elevated renal index,creatinine,serum uric acid,and urea nitrogen,and markedly decreased serum NO compared with the control group.Following intervention with phillyrin,there was a distinct decrease in serum uric acid,and renal function and histological morphology revealed varying degrees of recovery.Pathologi-cal damage in renal tissue was alleviated,and serum NO markedly increased.Conclusion Phillyrin exhibits a pronounced hypouricemic effect and remarkably alleviates renal damage in HUA mice induced by hypoxanthine combined with potassium oxonate,possibly through the improvement of endothelial dysfunction.关键词
