双硫死亡关键调控基因TCGA/GTEx联合队列分析揭示HNSCC预后生物标志物及潜在治疗靶点
Key Regulatory Genes of Disulfidptosis in a TCGA/GTEx Combined Cohort Reveals Prognostic Biomarkers and Potential Therapeutic Targets for Head and Neck Squamous Cell Carcinoma

Abstract
Objective To systematically analyze the prognostic value and immunoregulatory mechanisms of disulfidptosis-re-lated long non-coding RNAs(DRLs)in head and neck squamous cell carcinoma(HNSCC).Methods An integrated anal-ysis was performed on 511 HNSCC samples from TCGA and 44 normal tissues from GTEx.DRLs were screened using Pear-son correlation analysis(|R|>0.4,P<0.001),followed by univariate COX regression(P<0.05)and LASSO-COX regres-sion(10-fold cross-validation)to construct a prognostic model.Model performance was evaluated by Kaplan-Meier analy-sis,ROC curves(1/3/5-year AUC),and PCA.Immune microenvironment was assessed via ssGSEA,ESTIMATE,and TIDE analysis.RT-PCR was used to validate the expression of DRLs.Results A prognostic model incorporating six DRLs(AC083967.1,AC106820.5,etc.)was successfully established(3-year AUC=0.716).The high-risk group exhibited de-creased immune scores(P<0.05)and elevated TIDE scores(P<0.05),indicating a tendency for immune evasion.Func-tional enrichment analysis revealed that DRLs were involved in antigen presentation and NOD-like receptor pathways(FDR<0.05).Experimental validation confirmed differential expression of all six DRLs in HNSCC cell lines(vs.HOK,P<0.05).Conclusion DRLs serve as promising prognostic biomarkers for HNSCC and can regulate the immune microenviron-ment,offering novel targets for personalized therapy.关键词
