Detection and Genetic Evolution Analysis of PCV3 Pathogen on Core Genetic Swine Farms in Sichuan Province in 2024

Abstract

In order to identify the characteristics of epidemiology and molecular genetic variation of porcine circovirus 3(PCV3)in genetic pig population in Sichuan Province,2 976 clinical samples from pigs dead of diseases collected from core genetic swine farms in 8 regions of the province in 2024 were detected for PCV3 using real-time fluorescence quantitative PCR assay.The ORF2 genes of positive samples were amplified and sequenced,followed by genetic variation analysis for the sequences.The results showed that PCV3 was detected on all the farms,with an overall positive detection rate of 12.87%(383/2 976),which was the highest in Ya'an,reaching 52.51%(73/139),but the lowest in Mianyang,remaining at 0.63%(5/789).9 sequences of PCV3 ORF2 gene were obtained,all of which fell into PCV3a subtype,the Cap protein amino acid sequences deduced by the sequences were 94.4%-100%identical to those of 19 PCV3 strains prevalent in Sichuan Province from 2019 to 2023,and 98.5%-100%to those of 19 PCV3 strains isolated from origin countries including the United States,Canada and Denmark;compared to PCV3-US-MO2015,the classical strain,7 amino acid variation sites(S44C,Q55R,S77T,A97T,E128D,L150I and A152G)were available in the major antigenic epitope regions of the 7 sequences of Cap protein amino acid.In conclusion,PCV3 was widely prevalent in pig population in Sichuan Province,dominated by PCV3a subtype;pathogenicity and antigenicity of the virus might be affected by the mutations in the Cap protein amino acid sequences of the prevalent strains that primarily occurred in antigenic epitope regions.The subtypes and rules of genetic variation of PCV3 prevalent in Sichuan Province were revealed by the experiment,providing an important theoretical basis for future comprehensive prevention and control of PCV3 infection in the region.
