Effect of Glutamine Intervention on Monkeypox Virus Replication

Abstract
Monkeypox virus(MPXV)can exploit host metabolic pathways to enhance its replication.To clarify the role of the host metabolite glutamine(Gln)in MPXV replication,this study evaluated the effects of Gln intervention using quantitative real-time polymerase chain reaction(qPCR),plaque assays,and Western blot analysis to assess viral load,viral titers,and expression levels of viral proteins A5L and A29L.Additionally,Western blot was used to examine the effect of MPXV infection on the expression of glutamate dehydrogenase 1(GLUD1),a key protein in the Gln metabolic pathway.The results showed that,compared with the Gln+MPXV+ group,the Gln-MPXV+ group exhibited a significant reduction in MPXV viral load(P<0.000 1),viral titer(P<0.01),and expression of A5L and A29L proteins(P<0.000 1 or P<0.01).In contrast,the Gln++MPXV+ group showed no significant change in these parameters(P>0.05),indicating that Gln deprivation significantly inhibits MPXV replication,while Gln supplementation does not enhance it.Moreover,MPXV infection markedly suppressed GLUD1 protein expression(P<0.05),suggesting that GLUD1 may be a potential target through which MPXV modulates host glutamine metabolism.These findings indicate that blocking glutamine uptake may be a promising strategy for MPXV prevention and treatment,and provide a theoretical basis for the development of more effective antiviral approaches.关键词
