湖南省中部和西部地区山羊源肝片吸虫的遗传多样性和遗传进化分析
Genetic Diversity and Evolutionary Analysis of Fasciola hepatica Collected from Goats in the Central and Western Regions of Hunan Province

Abstract
To explore the genetic diversity of Fasciola hepatica(F.hepatica)isolated from goats in central and western Hunan Province and to analyze the characteristics of their ribosomal and mitochondrial gene sequences as well as their genetic evolutionary relationships,this study examined 36 goat-derived F.hepatica isolates from Zhangjiajie,Xiangxi,Huaihua,Shaoyang,Yongzhou,and Loudi.PCR amplification and sequencing were performed for five genetic loci:18S ribosomal RNA(18S rRNA),internal transcribed spacer 1(ITS-1),internal transcribed spacer 2(ITS-2),mitochondrial cytochrome C oxidase subunit I(cox1),and NADH dehydrogenase subunit I(nad1).Sequence characteristics,intraspecific and interspecific differences,and phylogenetic relationships were analyzed.The results showed that the sequence lengths for 18S rRNA,ITS-1,ITS-2,cox1,and nad1 genes were 585,453,360,676,and 903 bp,respectively.Among these,18S rRNA and ITS-1 exhibited a GC base preference,while ITS-2,cox1,and nad1 were AT-rich.The intraspecific variations for these genes were 0-1.0%,0-2.0%,0-0.3%,0-2.7%,and 0-1.7%,respectively,while interspecific differences ranged from 0.68%-1.85%,0.44%-3.52%,1.11%-9.44%,1.48%-13.46%,and 8.31%-14.51%,respectively.Phylogenetic analysis revealed that all 36 isolates clustered on the same branch,indicating that despite low levels of genetic diversity among goat-derived F.hepatica in these regions,the isolates share close genetic relationships.Additionally,ITS-2 and nad1 genes were identified as useful genetic markers for interspecific studies of Fasciola species.关键词
