Abstract
To analyze the epidemiological characteristics and genetic variation of porcine reproductive and respiratory syndrome virus(PRRSV)in Beijing from 2020 to 2024,a total of 6 127 clinical samples were collected from pig farms of various scales across eight districts in Beijing.Real-time fluorescent quantitative reverse transcription PCR(RT-qPCR)was used to detect PRRSV infection.The open reading frame 5(ORF5)of PRRSV was amplified by PCR,and homology and phylogenetic analyses were conducted based on the ORF5 gene.The results showed an overall PRRSV positivity rate of 16.35%(1 002/6 127)during 2020 to 2024,with a declining trend in detection rates over the years.The regional detection rates ranged from 10.31%to 22.84%,with the highest rate observed in Yanqing District.Detection rates varied among sample types,with the highest in lung samples at 22.54%(222/985),and the lowest in environmental samples at 3.72%(16/430);PRRSV was not detected in boar semen samples.A total of 26 PRRSV ORF5 gene sequences were obtained through sequencing.Phylogenetic analysis revealed that the PRRSV strains detected in Beijing mainly belonged to lineage 1.8(NADC30-like,42.31%,11/26)and lineage 1.5(NADC34-like,34.62%,9/26),while a few strains belonged to lineage 5(VR2332-like)and lineage 8(HP-PRRSV-like).This study reveals the epidemic characteristics and genetic diversity of PRRSV-2 in Beijing,providing a scientific basis for the development of targeted prevention and control measures.关键词
