龙胆泻肝汤对实验性自身免疫性葡萄膜炎模型小鼠的影响
Effect of Longdan Xiegan Decoction on Experimental Autoimmune Uveitis in Model Mice

Abstract
Objective To investigate the effect of Longdan Xiegan Decoction on experimental autoimmune uveitis(EAU)model mice and its mechanism.Methods The EAU model mice were reproduced by subcutaneous injection of 50 µL lymph fluid(interphotoreceptor retinoid-binding protein-Freund's complete adjuvant,1∶1,V/V)at each site(head,neck,bilateral hind leg roots,and tail roots of mice)and intraperitoneal injection of 1 µg pertussis toxin.A total of 48 model mice were randomly divided into model group(equal volume physiological saline),Longdan Xiegan Decoction group(1 mg/g),Longdan Xiegan Decoction+lipopolysaccharide(LPS)group(1 mg/g+1 µg),and LPS group(1 µg),with 12 mice in each group.The corresponding drugs or distilled water were administered by gavage once a day for 15 d.After the last gavage,1 µg LPS was injected by caudal vein at each time.Another 12 healthy mice were taken as the control group(equal volume of distilled water).The ocular inflammation of mice was observed under slit-lamp microscope and the clinical symptoms score was performed.Hematoxylin-eosin staining was used to observe the histopathology of mouse retina and score.The levels of inflammatory cytokines in serum and eye tissue were detected by enzyme linked immunosorbent assay.The proportion of type 1 helper T cells(Th1)/type 2 helper T cells(Th2)was analyzed by flow cytometry.The expression level of Toll-like receptor 4(TLR4)/nuclear factor-κB(NF-κB)signaling pathway related proteins was detected by Western blot.Results Compared with the model group,the clinical symptom score and retinal histopathological score in the Longdan Xiegan Decoction group were significantly decreased(P<0.05);the levels of interleukin 6(IL-6),IL-17,tumor necrosis factor-α(TNF-α)and interferon-γ(IFN-γ)in serum and eye tissue were significantly decreased,while the levels of IL-4 and IL-10 were significantly increased(P<0.05);the percentage of Th1 cells and the expression levels of TLR4,p-p65/p65,p-IκBα/IκBα in eye tissue were significantly decreased,while the percentage of Th2 cells was significantly increased(P<0.05).Compared with LPS group,the change trend of above indexes in Longdan Xiegan Decoction+LPS group was consistent.Conclusion Longdan Xiegan Decoction can reduce the ocular and peripheral inflammatory reactions and restore the immune homeostasis of Th1/Th2 in EAU model mice,which may be related to the inhibition of TLR4/NF-κB signaling pathway activation.关键词
