活血潜阳祛痰方改善肥胖高血压大鼠心肌细胞焦亡的机制研究
Mechanistic study on Huoxue Qianyang Qutan Recipe in alleviating cardiomyocyte pyroptosis in obesity hypertension

Abstract
Objective:To observe the effects and potential mechanisms of the Huoxue Qianyang Qutan Recipe(HQQR)on cardiomyocyte pyroptosis in obesity hypertension(OBH).Methods:An OBH rat model was established by feeding spontaneously hypertensive rats(SHR)with a high-fat diet.Successfully modeled OBH rats were randomly divided into the model group(OBH),valsartan group(Valsartan),low-dose HQQR group[HQQR(L)],and high-dose HQQR group[HQQR(H)].Drug interventions were administered for 10 weeks.WKY and SHR rats served as the control groups.Primary cardiomyocytes(CMs)were isolated and cultured from SD rats and treated with angiotensin Ⅱ(Ang Ⅱ)to establish a CMs injury model.The CMs were divided into the control group,Ang Ⅱ group,low-dose HQQR group[Ang Ⅱ+HQQR(L)],high-dose HQQR group[Ang Ⅱ+HQQR(H)],and valsartan group[Ang Ⅱ+Valsartan].ELISA was used to measure IL-1β and IL-18 levels in tissues and cell supernatants.Cell viability was assessed using the CCK-8 assay.Western blot was employed to detect the protein expression of NOD-like receptor protein 3(NLRP3),Caspase 1/p10,and the N-terminal domain of gasdermin D(GSDMD-N).Statistical analysis was performed on the related data.Results:In OBH rat myocardial tissues,IL-1β and IL-18 levels were significantly elevated(P<0.001),and the expression of NLRP3,Caspase 1/P10,and GSDMD-N proteins was significantly increased(P<0.001).HQQR treatment significantly reduced myocardial IL-1β and IL-18 levels(P<0.001)and decreased NLRP3,Caspase 1/P10,and GSDMD-N protein expression(P<0.00l).Ang Ⅱ stimulation significantly reduced CMs viability,increased IL-1β and IL-18 levels in cell supernatants compared to the blank group(P<0.001),and elevated NLRP3,Caspase 1/P10,and GSDMD-N protein expression(P<0.001).HQQR intervention improved CMs viability(P<0.05),reduced IL-1β and IL-18 levels(P<0.001),and significantly decreased NLRP3,Caspase 1/P10,and GSDMD-N protein expression(P<0.001).Conclusion:HQQR may improve CMs injury by inhibiting inflammation and reducing pyroptosis.关键词
