基于R值综合评价的金水缓纤方干预肺纤维化小鼠的作用特征分析
Analysis of characteristics on Jinshui Huanxian Formula treating mice with pulmonary fibrosis based on R-value comprehensive evaluation
摘要
Abstract
Objective:To observe the efficacy of Jinshui Huanxian Formula on different stages of multiple bleomycin-induced pulmonary fibrosis in mice.Methods:Ninty-six C57BL/6J mice were randomly divided into Control group,Model group,Jinshui Huanxian Formula(JHF)group,and Pirfenidone(PFD)group,with 3 batches(8 mice/batch)in each group.The PF mice were prepared by intratracheal injection of 3 mg/kg bleomycin solution on the 1st,14th and 28th day,respectively,and the mice in each group were gavaged with the corresponding drugs on the days 28-42,42-56 and 56-70,and the samples were collected on the 42nd,56th,70th day,respectively.The final body weight,lung function,lung coefficient,lung histopathology,collagen deposition,inflammation level,oxidative stress and other indexes were detected in each group of mice,and evaluated by R-value comprehensive evaluation method.Results:For lung function,lung function decreased in PF mice at all stages(VC,Cdyn,Cchord decreased)(P＜0.05,P＜0.01),and JHF significantly improved lung function on the 42nd and 56th day(P＜0.05,P＜0.01);for collagen deposition,collagen levels increased in PF mice at all stages(Fibrosis score,Col-Ⅰ and Col-Ⅲ increased)(P＜0.05,P＜0.01),and JHF improved collagen deposition at all stages(P＜0.05,P＜0.01);in terms of inflammation,inflammation levels were increased in PF mice at all stages(alveolitis scores,BALF cell counts,IL-6,IL-1β,TNF-α increased)(P＜0.05,P＜0.01),and JHF significantly reduce the level of inflammation on the 56th day(P＜0.05);in terms of oxidative stress,the oxidative stress level was elevated in PF mice at all stages(elevated MDA,decceased T-SOD,GSH)(P＜0.05,P＜0.01),and JHF significantly improved the level of oxidative stress on the 42nd day(P＜0.01),and the effect was better than that of PFD group.Conclusion:Jinshui Huanxian Formula has good efficacy on different stages of pulmonary fibrosis in mice.It can improve lung function,reduce collagen deposition and oxidative stress in inflammatory stage;reduce lung function,collagen deposition and inflammatory reaction in fibrotic formation stage;improve collagen deposition in fibrotic stage;and its efficacy is better than that of Pirfenidone in inflammatory stage.关键词
