高效液相色谱法测定茶叶中12种有机酸的研究
Study on the Determination of 12 Organic Acids in Tea by High Performance Liquid Chromatography

Abstract
The contents of tartaric acid,oxalic acid,formic acid,quinic acid,L-malic acid,pyruvic acid,acetic acid,lactic acid,fumaric acid,citric acid,succinic acid,and salicylic acid in tea were simultaneously determined by high performance liquid chromatography.The tea sample was extracted in a boiling water bath for 45 minutes,then filtered under reduced pressure while still hot and adjusted to volume.It was then filtered through a 0.22 μm aqueous filter membrane and analyzed by high-performance liquid chromatography using an AcclaimTM 120 C18 liquid chromatography column.Gradient elution using methanol as mobile phase A and 0.1 mol/L KH2PO4 as mobile phase B.The diode array detector detected the sample at a wavelength of 210 nm with an injection volume of 30 μL.The linear correlation coefficients of 12 organic acids were all above 0.999.The detection limits of 12 organic acids were 0.001~0.300 mg/g,the quantification limits were 0.003~1.000 mg/g,and the recovery rates were 91.8%~103.5%.The precision and re-peatability of the method were good.The results of the determination of six major types of tea and jasmine tea show that the major organic acids in the tested tea samples were mainly L-malic acid,quinic acid,and acetic acid,with content ranges of 4.70~30.25,8.36~25.75 and 3.98~28.80 mg/g,respectively.The study provided a reference for the de-velopment of standards for detecting organic acid components in tea.关键词
