Development of KASP Markers for the Whole Genome of Populus deltoides and Establishment of Elite Fingerprints

Abstract
The Poplar is an important tree species in artificial forests,yet it is difficult to accurately distinguish poplar varieties based on their morphology.To accurately identify superior varieties of Populus deltoides,KASP primers for the entire genome of Populus deltoides were developed based on the resequencing data of 95 natural populations of Populus deltoides.After alignment analysis,a total of 1 948 815 SNPs were obtained from the entire genome of Populus deltoides.A total of 48 125 loci with a distance of no less than 10 kb between adjacent SNPs were screened out,and KASP prim-ers were successfully designed for 25 478 of these loci.Ten groups of KASP markers were randomly selected from each chromosome and aligned with the reference genome of Populus simonii to obtain 44 KASP markers that were commonly used in Populus simonii.Using 10 superior varieties of Popu-lus deltoides,the above 44 groups of KASP markers were screened.Eventually,18 KASP markers with clear typing and high polymorphism were selected,with an average MAF of 0.34,an average ob-served heterozygosity of 0.45,an average polymorphism information content of 0.44,and the number of genotyping deletion samples ≤1.Genotyping was performed on 36 Populus deltoides samples using 18 groups of KASP markers screened,and the DNA fingerprint profiles of 36 poplar trees were successfully constructed.Among the 34 varieties of Populus deltoides,there were an average of 10.83 SNP locus differences in every two germplasms.Eventually,8 core KASP markers were screened out,which could accurately distinguish the 34 varieties of Populus deltoides.关键词
