Research on Physiological Response to Water-soaking Disorder of Citrullus lanatus During Ripening

Abstract
To explore the physiological response mechanism of flesh water infiltration damage during the ripening process of Citrullus lanatus,Citrullus lanatus germplasms 07058(resistant)and Yixuan(easy)with significant differences in flesh water infiltration were used as materials.The proportion of flesh damage,the content of glycolic acid,the contents of pyruvate(PA)and malondialdehyde(MDA),the activities of antioxidant enzymes such as SOD,CAT and POD,the contents of cell wall structural substances such as pectin and cellulose,and the activities of cell wall degrading enzymes were determined at five stages during the ripening process of Citrullus lanatus.The results showed that 20 days after pollination,the proportion of easily water-soluble material in the pulp rose rapidly.35 days after pollination,the damage proportion reached about 45%,while the water-resistant mate-rial 07058 showed no obvious water-soluble phenomenon.Compared with the water-resistant pickling material 07058 of the same period,the contents of sucrose,glucose and pulp acid in Yixuan were all higher than those in 07058 20 days after pollination,and the fructose content first increased and then decreased,which was lower than that in 07058 25 days after pollination.The contents of PA and MDA rose rapidly and were significantly higher than those of 07058.At the initial stage of waterlog-ging in Yixuan(20 days after pollination),the activities of SOD and CAT increased rapidly,higher than 07058,and then decreased rapidly,lower than 07058.The POD activity first declined and then rebounded,but the activity in each period was lower than 07058.The cellulose content and total pec-tin content of Yixuan were generally lower than those of 07058,while the soluble pectin content was higher than that of 07058.During the rapid water-soaking stage of the pulp(20 to 25 days after polli-nation),the cellulase(CL)and pectin lyase(PL)activities of Yixuan rose rapidly and were higher than those of 07058.At 25 days after pollination,the activity of Yixuan PL was 1.9 times that of 07058.The results of the correlation analysis showed that the waterlogging damage of Citrullus lana-tus was significantly positively correlated with multiple physiological indicators such as sucrose con-tent,MDA content,and CL activity.It was significantly negatively correlated with fructose content,SOD activity and cellulose content,etc.Studies had shown that waterlogging of Citrullus lanatus flesh was closely related to multiple physiological activities such as sugar metabolism,membrane damage,oxidative stress response,and cell wall degradation,and was the result of the coordinated changes in multiple physiological processes.关键词
