Abstract

To develop Artemisia argyi specific SSR molecular markers for constructing germplasm re-source fingerprint maps and variety identification,based on the previous Artemisia argyi transcrip-tome sequencing data,SSR site discovery was carried out using MicroSAtellite software,Primer 3.0 batch primers were designed,and capillary electrophoresis sequencing analysis was conducted after PCR amplification.The results showed that 21 980 SSR loci were detected,distributed across 20 069 transcripts,and 15 961 pairs of SSR primers were designed in batches.Among the 30 randomly se-lected pairs of primers,10 pairs with high polymorphism were screened out.Further,specific SSR primer combinations for constructing fingerprint maps of 25 Artemisia argyi germplasm resources and identifying the four famous Artemisia argyi and Nanyang Artemisia argyi series varieties were screened out.The transcriptome of Artemisia argyi was rich in SSR molecular marker sites.The de-velopment of Artemisia argyi specific SSR molecular markers could provide strong support for re-search such as genetic diversity analysis,fingerprint map construction,molecular-assisted breeding,and variety identification of Artemisia argyi.
