摘要

Abstract

To explore the genetic diversity of Artemisia argyi and its genetic differentiation from its close relative Artemisia anomala,Based on the ITS and matK sequences of 16 Artemisia argyi germ-plasms and 1 Artemisia anomala,the sequence characteristics,distances and phylogeny were ana-lyzed using Genious11.0 and MEGA11.0,etc.The results showed that the conserved length of the ITS sequences of 16 Artemisia argyi germplasms was 225 bp,the GC content ranged from 55.11%to 59.44%,and there were 3 base substitutions.The matK length was 723 bp to 724 bp,the GC content was 34.58%to 34.85%,and it contained 2 base substitutions and 1 base insertion.The ITS and matK sequences of Artemisia anomala were compared and analyzed with 16 WTBXArtemisia argyi germ-plasms.It was found that the ITS length of Artemisia anomala was 225 bp,the GC content was 56.44%,and there were 12 base substitutions.The matK length was 760 bp,the GC content was 34.56%,and it contained 2 base substitutions and 2 base insertions.The genetic distances of ITS and matK in 16 Artemisia argyi germplasms were 0-0.014 and 0-0.003 respectively,and their genetic distances were relatively close.The genetic distances between Artemisia anomala and 16 Artemisia argyi germplasms were 0.043-0.059 and 0-0.004 respectively,indicating a relatively long genetic distance.Phylogenetic tree analysis indicated that the ITS sequence divided 16 Artemisia argyi germ-plasms and Artemisia anomala into two major lineages,while the matK sequence failed to distinguish between Artemisia argyi and Artemisia anomala.Studies had shown that ITS sequences,due to their higher rate of variation and haplotype diversity,were more effective in analyzing intraspecific ge-netic diversity and distinguishing closely related species(such as Artemisia argyi and Artemisia anomala).The highly conserved matK sequence provided less information on intraspecific or interspe-cific differentiation and was more suitable for higher-order phylogenetic reconstruction.
