基于TLRs/MyD88/NF-κB信号通路探讨镇心安神方对特应性皮炎小鼠皮肤免疫屏障的作用及机制
Effects and Mechanisms of Zhenxin Anshen Formula on Skin Immune Barrier in Atopic Dermatitis Mice Based on the TLRs/MyD88/NF-κB Signaling Pathway

Abstract
Objective:To investigate the effects and mechanisms of Zhenxin Anshen Formula on the skin immune barrier in atopic dermatitis(AD)model mice based on the TLRs/MyD88/NF-κB signaling pathway.Methods:A total of 60 BALB/c mice were randomly divided into six groups:blank group,model group,loratadine+hydrocortisone butyrate group,and Zhenxin Anshen Formula high,medium,and low dose groups,with 10 mice in each group.The atopic dermatitis mouse model was established by 1-chloro-2,4-dinitrobenzene(DNCB)induction.Mice were treated with oral administration once daily for 15 consecutive days.At the end of the treatment,the following assessments were performed:30-minute scratching counts,skin lesion scoring,pathological examination of skin lesions,and detection of immunoglobulin E(IgE)levels by ELISA.The expression levels of TLR2,TLR4,and MyD88 proteins in skin lesions were evaluated by Western Blot.Results:Compared to the blank group,the model group showed significantly increased 30-minute scratching counts(P<0.01),higher skin lesion scores(P<0.01),and severe pathological damage in skin tissues,including large-scale epidermal necrosis and shedding,nuclear fragmentation,dermal necrosis,connective tissue proliferation,neutrophil infiltration,hair follicle destruction,and structural disorganization.Serum IgE levels and TLR2,TLR4,and MyD88 protein expression levels were significantly increased(P<0.05).Compared to the model group,the Zhenxin Anshen Formula groups exhibited significantly reduced 30-minute scratching counts(P<0.05),lower skin lesion scores(P<0.05,P<0.01),and significant improvement in skin histopathology with intact epidermal structure,orderly collagen fibers in the dermis,and normal distribution of skin appendages such as hair follicles and sebaceous glands.Serum IgE levels and TLR2,TLR4,and MyD88 protein expression levels were significantly decreased(P<0.05).Conclusion:Zhenxin Anshen Formula allebased ontes DNCB-induced pathological skin damage in AD mice,possibly by inhibiting TLR2,TLR4,and MyD88 protein expression,reducing serum IgE levels,and modulating immune response.These effects improve skin inflammation and itching,thereby protecting the skin immune barrier.关键词
