Abstract

Objective:This experiment employed iTRAQ proteomics technology to investigate age-related changes in mouse liver mitochondria using 3-,6-,and 12-month-old mice as experimental subjects.By comparing protein expression patterns in liver mitochondria among these age groups,we aimed to identify protein biomarkers closely associated with aging and examine the effects of Sijunzi Decoction on these biomarkers.The study sought to elucidate the molecular mechanisms of aging and the biological basis of Sijunzi Decoction's intervention in age-related syndromes from a proteomics perspective,thereby providing scientific evidence for formula-syndrome research.Methods:Fifty naturally aging mice of different ages were selected as experimental subjects,including ten 3-month-old mice and forty mice that naturally aged to 6 and 12 months through feeding.The mice were divided into groups according to experimental requirements:the 3-month-old mice served as the blank group,while the 6-and 12-month-old mice were randomly divided into two groups-the model group(20 mice)and the Sijunzi Decoction group(20 mice).During the experiment,the Sijunzi Decoction group received intragastric administration at a dose of 9.75 g·kg-1·d-1,while the blank and model groups received an equal volume of saline.On day 30 of the experiment,mouse livers were extracted and stored at-80℃for subsequent analysis.Proteomics analysis methods were employed to examine the liver samples.Results:Comparative analysis identified 734 differentially expressed proteins between the blank group and 6-month model group,and 639 differentially expressed proteins between the blank group and 12-month model group.The Sijunzi Decoction treatment induced beneficial regulation of 515 characteristic proteins in the 6-month model group and 389 characteristic proteins in the 12-month model group.Intersection analysis revealed 193 differentially expressed proteins that were regulated by Sijunzi Decoction in both age groups.Conclusion:The proteomics study demonstrated that Sijunzi Decoction significantly regulates the PPAR signaling pathway and Huntington's disease-related proteins in 6-month-old mouse liver tissue,while showing regulatory effects on Alzheimer's disease and Parkinson's disease pathways in 12-month-old mouse liver tissue.Sijunzi Decoction may exert its protective and regulatory effects on age-related physiological dysfunction by modulating key proteins in these signaling pathways.
