针刺对CUMS抑郁大鼠肠内容物代谢水平的调节
Regulation of Acupuncture on Metabolic Levels of Intestinal Contents in CUMS Depressive Rats

Abstract
Objective:To investigate the changes in metabolic levels of intestinal contents in rats with chronic unpredictable mild stress(CUMS)-induced depression before and after acupuncture treatment,and to elucidate the mechanism of acupuncture in treating depression at the intestinal level.Methods:Rats were randomly divided into a control group,a CUMS group,and an acupuncture group.The control group received no special treatment except normal food and water.The CUMS group and the acupuncture group were first subjected to model establishment.After successful modeling,the acupuncture group received acupuncture treatment.During the treatment period,a sucrose consumption test was conducted at fixed times each week.After the treatment,an open field test was performed.Finally,the intestinal contents were analyzed using GC-MS metabolomics.Results:After modeling,compared with the control group,the sucrose consumption of the CUMS group was significantly reduced(P<0.01).In the first two weeks of acupuncture treatment,the sucrose consumption of the acupuncture group showed a steady increase but had not yet reached the level of recovery.Compared with the control group,the difference was statistically significant(P<0.01).After the third week of acupuncture treatment,the sucrose consumption of the acupuncture group returned to the level of the control group,with no statistically significant difference between the two groups(P>0.05).There was no significant difference in the total movement distance(P>0.05).Compared with the control group,the central movement time of the rats after acupuncture treatment showed no significant difference(P>0.05).The intestinal contents of the CUMS group,including spermine,ethanolamine,L-tyrosine,L-homoserine,β-alanine,L-histidine,L-alanine,3,4-dihydroxyphenylbutyric acid,2-pyrocatechuic acid,syringic acid,trehalose,α-D-glucose,D-arabinose,oxalic acid,and phenylacetic acid,were reversed after three weeks of acupuncture treatment.Conclusion:Acupuncture can reverse anhedonia and depressive-like behaviors in CUMS rats,preliminarily confirming the efficacy of acupuncture in treating depression.The differential metabolites reversed by acupuncture are mainly involved in the following metabolic pathways:β-alanine metabolism,histidine metabolism,glyoxylate metabolism,and dicarboxylate metabolism.关键词
