马度米星铵对小鼠毒性及毒理学机制研究
Toxicity and toxicological mechanism of madumeicin ammonium in mice

Abstract
The aim was to investigate the toxic effects of Madomicin ammonium on mice and to understand its toxic target organs.Feeding 60 Kunming mice with SPF grade was randomLy divided into 6 groups with 10 mice in each group(half male and half female),the control group was given olive oil,and the drug groups 1-5 were given malduamicin ammonium by single intragastric administration with concentrations of 2.500,1.768,1.250,0.884 and 0.625 mg/mL,and the administration volume was 20 mL/kg,respectively.After 14 days of continuous observation,the physical signs,poisoning and death status of the mice were recorded.The dead mice were dissected,the organ changes were observed in general,and the fixed tissue samples were sliced to observe the pathological changes.The coherent protein expression of NF-κB/TLR-4/JNK signaling pathway was detected by Western Blot assay in liver and lung.Haematology indexes showed that there were significant differences between blank group and drug administration group(P<0.05).All the mice in each administration group were depressed and inactive after administration,among which the dead mice could be seen gastrointestinal distension,yellow and fragile liver,and white lung.All the surviving mice recovered to normal on the 7th day,and no abnormalities were found in the anatomy on the 15th day.Histopathology showed that the liver underwent degeneration and necrosis in different degrees,the alveolar wall was thickened,the alveolar cavity collapsed and narrowed,and some renal tubule epithelial cells were swollen.Western Blot analysis showed that the relative expressions of TLR4,NF-κB P-p65,NF-κB p65,MyD88 and JNK in liver were significantly increased(P<0.01).Studies have shown that madomicin is highly toxic to mice.The LD50 was 38.49 mg/kg body weight.The 95%confidence limit for LD50 was 36.67-40.4 mg/kg.The main target organs of toxicity were liver and lung.关键词
