黑曲霉发酵常山条件优化及其对常山碱含量的影响
Optimization of fermentation conditions of Aspergillus niger in Changshan and its effect on the content of febrifugine

Abstract
In order to investigate the effect of Aspergillus niger fermentation on the content of febrifugine in Changshan,Aspergillus niger was used to ferment Changshan,with cellulase activity as the evaluation index,the optimum fermentation process was optimized by single factor investigation method and orthogonal test method.The fermented Changshan was prepared by the best fermentation process,and the total alkaloids of Changshan and fermented Changshan were prepared according to the method of preparing alkaloids.The content of febrifugine in Changshan and fermented Changshan was determined and compared by HPLC.The results showed that the optimal conditions for fermentation were:a temperature of 30℃,an inoculum volume of 2%,a moisture content amounting to 65%,and fermentation duration of 9 d.Given the optimal conditions mentioned,the cellulase activity attained a level of 121.67 U/g.The mass fraction of febrifugine in the fermented Changshan was 0.046%,and the mass fraction of febrifugine in the Changshan was 0.023%.The content of febrifugine in the fermented Changshan increased by 97.38%after Aspergillus niger fermentation.It shows that Aspergillus niger fermentation is helpful to improve the extraction rate of febrifugine in Changshan.The research results can provide a basis for the further development and utilization of Changshan.关键词
