Abstract

[Objective]To investigate the biological function and signal peptide characteristics of the maize Lazarus 1(LAZ1)proteins.[Meth-od]The transmembrane structure and signal peptides of nine members of the ZmLAZ1 family were analyzed using bioinformatic method.The signal peptide secretion function was verified using a pSUC2 vector in combination with the yeast strain YTK12,which has a sucrose invertase secretion defect.[Result]All members of the ZmLAZ1 family contain a high-probability transmembrane helix at the N-terminus.Among them,ZmLAZ1-2,ZmLAZ1-3,ZmLAZ1-4,ZmLAZ1-5,ZmLAZ1-7,and ZmLAZ1-9 were predicted to have signal peptides.Yeast secretion experiments further confirmed that the ZmLAZ1-7 signal peptide possesses secretion activity and the ZmLAZ1-7 signal peptide fusion protein were capable of secreting invertase,enabling the breakdown of raffinose into monosaccharides.[Conclusion]This study provides theoretical support for further elucidating the biological functions of ZmLAZ1 family proteins and their fictional mechanisms in maize,and provides a reference for the study of gene functions and breeding applications in maize.
