摘要

Abstract

[Objective]To investigate the evolutionary origin,subcellular localization,and functional role of the CGLD27 LIKE(AT5G11840)protein in carotenoid biosynthesis in Arabidopsis thaliana,as well as its influence on plant growth and photoprotection.[Method]The evolution-ary origin and phylogenetic distribution of CGLD27 LIKE were analyzed using bioinformatics tools.Transgenic plants overexpressing CGLD27 LIKE were generated through molecular biology techniques.Expression levels of genes involved in the carotenoid biosynthesis pathway and the accumulation of photosynthetic pigments in the transgenic lines were quantitatively assessed.[Result]CGLD27 LIKE is of cyanobacterial origin and is highly conserved across photosynthetic organisms.Subcellular localization analysis revealed its chloroplast localization.Overexpression of CGLD27 LIKE resulted in phenotypic alterations,including dwarfism,chlorosis,and reduced fertility.Photosynthetic pigment content was signifi-cantly decreased.The albino phenotype exhibited light-dependent characteristics:it was exacerbated under short-day conditions but partially al-leviated under continuous light exposure.qRT-PCR analysis demonstrated down-regulation of key rate-limiting enzyme-encoding genes such as PSY and its downstream gene LCYb,whereas PDS and ZDS were significantly up-regulated,indicating the presence of a feedback regulatory mechanism in carotenoid biosynthesis.[Conclusion]As an evolutionarily conserved chloroplast-localized protein,CGLD27 LIKE may act as a negative regulator of carotenoid biosynthesis by modulating PSY expression.Its overexpression disrupts photoprotective mechanisms and impairs plant development,thereby providing a novel genetic target for improving crop stress tolerance and nutritional quality.
