低剂量辐射调节肠道菌群改善2型糖尿病小鼠雄性生殖损伤的作用
Effects of Low-Dose Radiation on Gut Microbiota to Improve Male Reproduc-tive Damage in Type 2 Diabetes Mice

Abstract
Objective To explore the effects of low dose radiation on gut microbiota in 2 type diabetes mice,and to investigate the role and mechanisms of alleviating male reproductive damage in diabetes.Methods Type 2 diabetes(T2DM)mouse model was established by feeding high-fat diet and injecting STZ.Eighty 80 male C57BL/6 mice were divided into control group(Con),low-dose irradiation group(LDR),type 2 diabetes model group(T2DM),and type 2 diabetic model+low-dose group(T2DM+LDR).The diabetic model was established by intraperitoneal injection of STZ at 30 mg/kg after feeding with high-fat diet for 4 months.After 14 rounds of 0.075 Gy LDR every other day,16S rDNA sequencing was used to detect the effects on the abundance and composition of gut microbiota;Hema-toxylin eosin staining(HE)was used to detect testicular tissue damage,while Western Blotting was used to detect TLR4,MyD88,and NF-κB expression.Resluts T2DM mouse model was successfully established,showing impaired glucose tolerance and elevated blood glucose levels,and the glucose level exceeded 11.1 mmol/L.As compared with the Con group,mice in the T2DM group showed a signifi-cant increase in body weight and blood glucose levels(P<0.05),and the β cell damage were also significantly increased in pancreatic is-lets of the mice,the seminiferous tubules were thickened in the testicles,exhibiting an inflammatory response.As compared with the T2DM group,the indicators described above were significantly improved in LDR+T2DM groups(P<0.05).The 16S rDNA sequencing re-sults showed a total of 308 and 225 characteristic OUTs in the T2DM and LDR+T2DM groups,indicating that LDR can reduce the compo-sition of gut microbiota in T2DM.In addition,the abundance of Bacilli decreased in T2DM mice,while the abundance of Desulfobacterota significantly increased.LDR can alleviate the decrease of Bacilli and the increase of Desulfobacterota in T2DM.In addition,WB results showed that TLR4,MyD88,and pNF-κB p65 expressions were increased in the testicular tissue of T2DM groups,while the protein ex-pression described as above were decreased in the LDR+T2DM group.The mRNA results showed that TLR4,MyD88,and NF-κB mRNAs were similar to the protein expressions,and as compared with the Con group,the TNF-α and IL-1 mRNAs increased in T2DM group,as compared with T2DM groups,the TNF-α and IL-1 mRNAs significantly decreased in mice of LDR+T2DM groups(P<0.05).Conclusion LDR can reduce the increase of Desulfobacterota,inhibit the TLR4/MyD88/NF-κB pathway and lower in flammation,thereby alleviating testicular injury caused by diabetes.关键词
