摘要

Abstract

Objective The extraction process of total flavonoids and polyphenols from Perilla frutescens leaves was optimized,and their in vitro hypoglycemic activity was evaluated.Methods Using single-factor and orthogonal experiments,the extraction process of total flavo-noids and polyphenols from P.frutescens leaves was optimized.The in vitro hypoglycemic activity of the extracts was evaluated by measur-ing the inhibitory effects on α-glucosidase activity.Results The optimal extraction condition for total flavonoids of P.frutescens leaves was as follows:ethanol concentration of 70%,material-liquid ratio of 1∶55(g∶mL),soaking time of 2 h,ultrasound time of 60 min,ultra-sound temperature of 60℃,and ultrasound power of 400 W.The content of total flavonoids in P.frutescens leaves was(290.30±2.50)mg/g.The optimal condition for total polyphenols of P.frutescens leaves was as follows:ethanol concentration of 40%,liquid-liquid ratio of 1∶35(g∶mL),soaking time of 3 h,ultrasound time of 40 min,and ultrasound power of 350 W.The total polyphenol content was(166.12±1.77)mg/g.The total flavonoids and polyphenol extracts of P.frutescens leaves inhibited α-glucosidase with IC50 values of(32.09±1.13)and(68.17±1.82)mg/L,respectively.Conclusion The optimized extraction process of total flavonoids and polyphenols from P.frutescens leaves was reproducible and reasonably feasible.The total flavonoids and polyphenols extracts of P.frutescens leaves showed good in vitro hypoglycemic activity,which provides reference for their development and utilization.
