UFLC-MS/MS法同时测定肾炎舒片中绿原酸和金丝桃苷的含量
Simultaneous Determination of Chlorogenic Acid and Hyperoside in Shenyanshu Tablet by UFLC-MS/MS

Abstract
Objective A method for simultaneous determination of chlorogenic acid and hypericin in Shenyanshu tablets by ultra-fast liq-uid chromatography tandem with mass spectrometry was established to provide reference for its quality control.Methods The separation was performed on Shim-Pack XR-ODS column(75 mm×3.0 mm,2.2 μm)with 0.1%formic acid aqueous solution(A)-acetonitrile(B)as mobile phase.Gradient elution(0-4 min,5%—12%B,4-13 min,12%—15%B,13-14 min,15%—80%B,14-15 min,80%—80%B)was conducted at a flow rate of 0.6 mL/min and a column temperature of 35℃.Detection was performed with electrospray ioniza-tion source in negative ionization mode.Quantification was performed using multiple reactions monitoring mode.Results The linear ranges of chlorogenic acid and hyperoside were 0.1-10 mg/L(r=0.998 3)and 0.01-1 mg/L(r=0.999 6).The average recoveries(n=9)were 99.2%and 98.7%.The content ranges of chlorogenic acid and hyperoside in 6 batches of samples were 0.063%-0.072%and 0.002 7%-0.003 3%,respectively.Conclusion The method has high sensitivity and good stability,and can be used for quality control of Shenyanshu tablets.关键词
