青蛤糖胺聚糖的分离及其体外抗氧化与降血糖活性评价
Isolation of glycosaminoglycan from Cyclina Sinensis and evaluation of its antioxidant and hypoglycemic activity in vitro

Abstract
Objective To isolate glycosaminoglycan from Cyclina sinensis and evaluate its antioxidant and hypoglycemic activities in vitro.Methods Glycosaminoglycan(S2)was extracted from soft parts of Cyclina sinensis by enzymolysis.The extraction efficiency of three enzymes(neutral protease,papain and trypsin)was investigated.Purified saminoglycan sample S2 was obtained by enzyme extraction and alcohol precipitation,Sevag method,anion exchange column chromatography and dialysis lyophilization.The content of glycosaminoglycan was determined by Alysin blue method,and the antioxidant activity of S2 was determined by measuring the clearance rate of ABTS(2,2'-diazine-bis-(3-ethylbenzodihydrothia-zoline-6-sulfonic acid)diammonium salt),hydroxyl free radical and DPPH free radical(1,1-diphenyl-2-trinitrophenylhydrazine).The inhibitory effects of S2 on α-glucosidase and α-amylase were determined to investigate its hypoglycemic activity.Results Neutral protease was identified as the optimal protease,with the yield and content of glycosaminoglycan in the enzymatic hydrolysate extract S1 being 0.14%and 10.48%,respectively.The purified sample S2 of glycosaminoglycan exhibited a content of 52.88%and demonstrated good antioxidant activity,with IC50 values for scavenging ABTS radicals,hydroxyl radicals,and DPPH radicals determined to be 0.435 mg/mL,1.590 mg/mL,and 3.724 mg/mL,respectively.Furthermore,S2 showed significant inhibitory effects on α-glucosidase and α-amylase,with IC50 values of 7.712 mg/mL and 0.671 mg/mL,respectively.Conclusion Glycosaminoglycan S2 exhibits good in vitro antioxidant and hypoglycemic activity,and has potential application value.关键词
