摘要

Abstract

Objective To study the differential chemical components of the Paeoniae Radix Alba(PRA)pieces processed with different moistening methods and drying temperatures,aiming to provide a reference for the processing technology of the PRA pieces.Methods The fingerprints of PRA and its different processed pieces with four moistening methods(direct water immersion,immersion followed by sealed moistening,immersion in 40℃warm water followed by sealed moistening,and steaming followed by moistening)and four drying temperatures(room temperature,40℃,50℃,and 60℃)were established by HPLC.The principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to analyze the differential components of PRA and its different processed pieces.Furthermore,the content changes of differential chemical components under different processing techniques were analyzed.Results The HPLC fingerprint similarity of PRA and its different processed pieces exceeded 0.900,and 16 common peaks were calibrated.PCA divided the different processed pieces by moistening and drying techniques into two categories.For moistening techniques,OPLS-DA screened 6 differential marker components.For drying techniques,4 differential components were screened.Analysis showed that the chemical constituents of glycosides and phenols were significantly related to the moistening temperature and duration,including gallic acid and 1,2,3,4,6-pentagalloylglucose.And the chemical constituents of glycosides were significantly related to the drying temperature,including benzoyl paeoniflorin and paeoniflorin.Conclusion When gallic acid,paeoniflorin,1,2,3,4,6-pentagalloylglucose,and benzoyl paeoniflorin were employed as indicator components,the results indicated that the optimal processing technology for PRA comprised immersing the raw materials until 60%-70%softened,followed by sealed moistening for uniform texture,slicing,and air-drying at room temperature.This study provides a valuable reference for the processing technology of the PRA pieces.
