Abstract

Objective To evaluate the detoxifying and liver-protective effects of Phyllanthus emblica L.(PE)on acute alcohol intoxication model rats,and explore its therapeutic mechanism.Methods Forty-two male SD rats were randomly divided into 7 groups,including normal control group,model group,high and low dose groups of PE fruit juice polyphenol extract,high and low dose groups of PE fruit residue polysaccharide extract,and positive control group of Gegen Zhiju compound tablets.In addition to the normal control group,the acute ethanol intoxication model was prepared by gavage of excessive alcohol 0.5 h after gavage of test samples of different concentrations in each experimental group.Serum biomarkers,including aspartate aminotransferase(AST),alanine aminotransferase(ALT),and ethanol levels,were quantified.Hepatic oxidative stress and metabolic parameters,including superoxide dismutase(SOD),malondialdehyde(MDA),glutathione(GSH),alcohol dehydrogenase(ADH),and aldehyde dehydrogenase(ALDH),were also determined.Histopathological assessment of gastric mucosal integrity was performed using hematoxylin and eosin(HE)staining.Results After administering excessive alcohol by gavage,the mental state and activity ability of rats in each treatment group were significantly better than those in the model group.The ethanol content of different groups treated with extracts of PE decreased significantly within 8 hours of drunkenness(P<0.05).The high-dose group of fruit polyphenol extract and fruit residue polysaccharide significantly reduced AST and ALT levels(P<0.01),and increased SOD,GSH,MDA,ADH,and ALDH levels in liver tissue(P<0.05 or P<0.01).Pathological sections of gastric mucosa tissue showed severe bleeding,extensive shedding of epithelial cells,severe tissue edema,and significant infiltration of inflammatory cells in the model group.All dose groups of PE extract improved the pathological changes of rat gastric mucosa tissue to varying degrees.Conclusion Extracts derived from fresh PE fruits can improve the activity of ethanol metabolizing enzymes,enhance antioxidant capacity,accelerate alcohol metabolism,and alleviate liver and stomach tissue damage,showing significant sobering,liver protective,and gastric mucosal protective effects on acute alcohol poisoning.
