咳嗽宁方治疗小鼠慢性咳嗽的作用及机制探讨
Effects and mechanism of Ke-sou-ning decoction in treating chronic cough in mice

Abstract
Objective To investigate the therapeutic effect of Ke-sou-ning decoction(KSND)on chronic cough(CC)in mice,and explore its mechanism of action by combining network pharmacology and experimental verification.Methods A CC mouse model was established by inhaling concentrated ammonia water to induce cough.After the model was established,low,medium,and high doses of KSND were administered for intervention treatment.The cough frequency and cough latency of mice were observed and recorded,and the latency extension rate and cough inhibition rate were calculated.HE staining was used to conduct pathological analysis of the trachea and lung tissues.The research method of network pharmacology was employed to screen the active ingredients,key targets,and signaling pathways of KSND for treating CC.The cytometric bead array(CBA)multi-factor flow cytometry kit was used to verify the predicted targets.Results KSND significantly reduced the cough frequency and prolonged the cough latency of CC mice,increased the latency extension rate and cough inhibition rate.It also restructured the damaged tracheal tissue of CC mice,restored the shape of alveoli,and alleviated inflammatory infiltration.The results of network pharmacology research showed that there were 164 potential core active ingredients and 309 potential targets and signaling pathways for KSND in treating CC,including AKT,TNF,STAT3,IL-10,etc.CBA results showed that compared to the model group,the expression level of TNF-α in the bronchoalveolar lavage fluid of mice in the treatment group was significantly reduced,while the expression level of IL-10 was significantly increased.Conclusion KSND can alleviate the symptoms of CC mice and reduce the pathological damage to lung and tracheal tissues,and its mechanism of action may be related to the regulation of inflammatory-immune balance.关键词
