摘要

Abstract

Objective To investigate the effect of Xiangsha Liujunzi Decoction on regulation of autophagy and apoptosis of Cajal interstitial cells(ICC)via the PERK/eIF2α pathway,providing experimental evidence for treating functional dyspepsia(FD).Methods FD rat models were induced with iodoacetamide and irregular diet,then divided into normal control(NC),model control(MC),and low-dose(LD),medium-dose(MD),high-dose(HD)Xiangsha Liujunzi Decoction groups.After 14 days of treatment,medicated serum was used to treat ICC cells.Gastric pathology was assessed by HE staining.Cell autophagy was measured by MDC and AO staining.Cell apoptosis was detected by flow cytometry.The expression of PERK,p-eIF2α,eIF2α was detected by western blot.Results MC group showed significant gastric damage,gland disarray,and inflammation,which were notably improved in the HD group.Cell autophagy was elevated in MC and reduced by HD.Cell apoptosis was increased in MC and decreased dose-dependently in LD,MD and HD groups(P<0.05).Western blot revealed PERK and p-eIF2α were decreased and eIF2α was increased compared with MC group.Treatment with Xiangsha Liujunzi Decoction restored PERK and p-eIF2α and reduced eIF2α.Conclusion Xiangsha Liujunzi Decoction inhibits ICC autophagy and apoptosis via the PERK/eIF2α pathway,improves gastrointestinal motility disorders in a dose-dependent manner,supporting FD treatment.
