Abstract
This review summarizes recent advances in the asymmetric synthesis of tapentadol,with a detailed focus on three key strategies:substrate-controlled induction,chiral auxiliary-assisted induction,and asymmetric catalysis.The design and optimization of synthetic routes based on these approaches are thoroughly discussed.By evaluating recently reported routes(within the last decade)against critical criteria such as stereoselectivity,reaction yield,and industrial feasibility,this work provides a comparative and critical analysis of various synthetic processes.The aim is to offer valuable insights and practical references for the efficient and environmentally sustainable industrial production of tapentadol.关键词
