摘要

Abstract

To determine the parasitic species collected in a fecal sample from a Debao pony(Equus caballus)under quarantine in a specific place in Guangdong Province,the collected parasite specimens were morphologically and molecular biologically identified.DNA was extracted from the parasites,and the internal transcribed spacer(ITS)sequence of the ribosomal DNA was amplified by PCR.Homology analysis was conducted through sequencing and BLAST.Furthermore,the complete mitochondrial genome of this worm was obtained by high-throughput sequencing,and its mitochondrial genome was analyzed for structural composition,base composition,tRNA and rRNA gene secondary structure.The results showed that the worms were preliminarily identified as strongylid nematodes by morphological observation and PCR amplification.The complete mitochondrial genome was 13 828 bp.The complete mitochondrial sequence was subjected to BLAST analysis to get the highest nucleotide sequence similarity of 95.20%with that of Cylicocyclus insigne,and the login number of the parasite's mitochondrial genome sequence was NC_013808.Genetic evolution analysis of mitochondrial genome sequences was conducted according to the sequences of 14 strongyloid nematode species from NCBI,the results showed that the worms were upon the branch of the Cylicocyclus auriculatum.It had the most close phylogenetic relationships with C.uriculatum.In summary,the worms from Debao pony fecal were identified as C.auriculatum based on the morphological observations and molecular biological methods,and the sequence analysis of complete mitochondrial genome was used to understand the genetic background of infected parasites,which laid the foundation for further improvement of the prevention and control measures against strongyloid nematodes in equines.
