Apelin-13调控自发性高血压大鼠心室重构的作用机制
Mechanism of Apelin-13 in regulating ventricular remodeling in sponta-neously hypertensive rats

Abstract
Objective To explore the effect of Apelin-13 on ventricular remodeling in spontaneously hypertensive rats and reveal its possible mechanism of action.Methods Spontaneously hypertensive(SHR)rats were randomly divided into 4 groups:the model group,the low-dose Apelin-13 group,the high-dose Apelin-13 group,and the Perindopril group,with 8 rats in each group.Another 8 WKY rats were set up as the normal control group.The cardiac function of rats was evaluated by echocardiography.Isolate the left ventricle of rats and calculate the cardiac mass index.Hematoxylin-eosin staining and Masson staining were used for pathological morphological analysis and fibrosis identification of rat myocardial tissue.The levels of inflammation-related cytokines such as tumor necrosis factor-α(TNF-α),interleukin(IL)-6,IL-1 and IL-1 βwere detected by enzyme-linked immunosorbent assay.Western blotting was used to detect the expression changes of proteins related to the nuclear factor-KB(NF-κB)/inducible nitric oxide synthase(iNOS)signaling pathway.Results The cardiac functions of rats in the low-dose Apelin-13 group,the high-dose Apelin-13 group and the Perindopril group were significantly stronger than those in the model group but weaker than those in the normal control group,and the differences were statistically significant(P<0.05).The levels of cardiac mass index and pro-inflammatory factors TNF-α,IL-6,and IL-1 β in the low-dose Apelin-13 group,high-dose Apelin-13 group,and Perindopril group were lower than those in the model group but higher than those in the normal control group,and the differences were statistically significant(P<0.05).The results of myocardial histopathological detection in the low-dose Apelin-13 group,the high-dose Apelin-13 group and the Perindopril group were significantly better than those in the model group,but slightly worse than those in the normal control group.The protein levels of NF-κB and iNOS in the low-dose Apelin-13 group,the high-dose Apelin-13 group and the perindopril group were lower than those in the model group but higher than those in the normal control group,and the differences were statistically significant(P<0.05).Conclusion Apelin-13 may have therapeutic ef-fects on ventricular remodeling and cardiac function in spontaneously hypertensive rats,and its mechanism may be related to the regulation of the NF-κB/iNOS pathway.关键词
