链脲佐菌素加膳食诱导的小鼠2型糖尿病模型研究
Study on the model of type 2 diabetes mellitus induced by streptozotocin and diet in mice

Abstract
Objective To establish the model of type 2 diabetes mellitus(T2DM)induced by streptozotocin(STZ)combined with diet in mice,and analyze the effects of drugs on laboratory indexes of the model.Methods Forty weaned male BALB/c mice were selected,of which 8 were fed with regular feed as the normal control group and 32 were fed with high-fat feed as the STZ model mice.The STZ model mice were divided into model control group,glibenclamide group,insulin group,and Rosiglitazone group by random number table method,with 8 mice in each group.The serum fasting blood glucose(FBG),triglycerides(TG),total cholesterol(TC),insulin,C-peptide,and blood glucose changes were compared between the groups.Results The serum FBG,TG,TC,and blood glucose levels at 0,30,60 and 120 min in the model control group were higher than those in the glibenclamide group,insulin group,Rosiglitazone group,and normal control group,the serum FBG,TG,TC,and blood glucose levels at 0,30,60 and 120 min in the glibenclamide group were higher than those in the insulin group,Rosiglitazone group,and normal control group,the serum FBG,TG,TC,and blood glucose levels at 0,30,60 and 120 min in the insulin group were higher than those in the Rosiglitazone group and normal control group,the differences were statistically significant(P<0.05).The serum insulin and C-peptide levels of the model control group were lower than those of the glibenclamide group,insulin group,Rosiglitazone group,and normal control group,the serum insulin and C-peptide levels of the glibenclamide group were lower than those of the insulin group,Rosiglitazone group,and normal control group,the serum insulin and C-peptide levels of the insulin group were lower than those of the Rosiglitazone group and normal control group,the serum insulin and C-peptide levels of the Rosiglitazone group were lower than those of the normal control group,and the differences were statistically significant(P<0.05).Conclusion In the T2DM rat model in-duced by STZ plus high-fat and high sugar diet,there was a significant increase in blood glucose,abnormal oral glucose tolerance,and insulin resistance,which were improved after drug treatment.Among them,the treatment effect of Rosiglitazone was the best.关键词
