Salt Tolerance and Physiological Response to NaCl Stress in Five Fraxinus chinensis Clones

Abstract
To study the differences in salt tolerance among different ash(Fraxinus chinensis)clones in saline and alkaline land,this study uses grafted seedlings of five clones(European Ash-Compact,European Ash-Open,12-62,12-76,and Yuanla 2)as test materials.This study simulates salt stress environments with NaCl solutions at different concentrations[1‰(CK),3‰,5‰ and 7‰],determines growth indicators,physiological and biochemical indicators and evaluates their salt tolerance.The results show that with the increasing NaCl concentration,the relative growth rate of all five ash clones decreases gradually,while biomass increases initially but decreases subsequently.The osmotic regulating substances like soluble sugar,soluble protein and free proline gradually increase.The chlorophyll content first increases and then decreases,reaching the maximum at 3‰ NaCl.The activity of superoxide dismutase(SOD)increases gradually,while the activity of peroxidase(POD)first increases and then decreases.The Na+/K+ratio increases extremely significantly.With the extension of stress time,the contents of osmotic regulating substances—soluble sugars,soluble proteins and free proline—increase significantly(P<0.05).The activities of SOD and POD enzymes first increase and then decrease,while the chlorophyll content decreases extremely significantly.Compared to the other four ash clones,clone 12-76 exhibits the highest relative seedling height growth rate,relative ground diameter increase rate,total biomass,and activities of SOD and POD enzymes,and the lowest malondialdehyde(MDA)content.A comprehensive evaluation of the growth,physiological,and biochemical indicators of different F.chinensis clones using the fuzzy membership function method shows that the salt tolerance ranking is:12-76>Yuanla 2>European Ash-Compact>European Ash-Open>Clone 12-62.关键词
