汉黄芩素减轻大鼠心肌缺血再灌注损伤的研究
Attenuation of Myocardial Ischemia/Reperfusion Injury in Rats by Wogonin

Abstract
Objective:To investigate the effects of wogonin(WOG)in attenuating myocardial ischemia/reperfusion injury(MI/RI)in rats by regulating the macrophage polarization via the phosphoinositide 3-kinase(PI3K)/protein kinase B(AKT)signaling path-way.Methods:Sprague Dawley(SD)rats were divided into Sham,Model,WOG low-dose(WOG-L),WOG high-dose(WOG-H),and WOG-H+LY294002(PI3K inhibitor)groups.Cardiac function,peripheral blood M1 and M2 macrophage levels,serum cardiac troponin I(cTnI),lactate dehydrogenase(LDH),tumor necrosis factor-alpha(TNF-α),interleukin-6(IL-6),and interleukin-10(IL-10)were measured.Myocardial histology and fibrosis were examined.Myocardial apoptosis,malondialdehyde(MDA)content,superoxide dismutase(SOD)and catalase(CAT)activities,and protein expression levels of Bax,Bcl-2,and PI3 K/AKT pathway components were assessed.Results:WOG treatment alleviated myocardial injury and fibrosis,improved left ventricular ejection frac-tion(LVEF)and left ventricular fractional shortening(LVFS),increased peripheral M2 macrophages and serum IL-10,enhanced myocardial SOD and CAT activities,and upregulated Bcl-2,p-PI3 K/PI3 K,and p-AKT/AKT protein expression.It also reduced pe-ripheral M1 macrophages,serum cTnI,LDH,TNF-α,IL-6 levels,myocardial apoptosis rate,MDA content,and Bax expression(P<0.05).The PI3K inhibitor LY294002 attenuated the protective effects of WOG on MI/RI rats(P<0.05).Conclusion:WOG may protect against myocardial injury in MI/RI rats by activating the PI3K/AKT signaling pathway.关键词
