Effects of Liqi Huatan Huoxue Formula-containing Serum on Mitochondria-mediated Cardiomyocyte Apoptosis

Abstract
Objective:To investigate the effects of Liqi Huatan Huoxue Formula-containing serum on mitochondria-mediated car-diomyocyte apoptosis.Methods:A co-culture model of primary rat cardiac microvascular endothelial cells(CMEC)and myocardial cells was established.The samples were randomly divided into the blank group,control group,siRNA interference group,Chinese medicine group(Liqi Huatan Huoxue Formula),siRNA+Chinese medicine group,nicorandil group,and valsartan group.Immuno-fluorescence staining was used to detect the degree of mitochondrial permeability transition pore(mPTP)opening.Fluorescent probes were used to assess mitochondrial membrane potential.Real-time quantitative PCR(qPCR)was applied to detect the relative mRNA expression levels of apoptosis-related factors in the mitochondrial pathway.Results:Compared with the blank group,the model group and siRNA interference group showed significantly increased relative mRNA expression levels of caspase-3,caspase-9,cytochrome c(Cyt-c),Bcl-2-associated death promoter(Bad),and Bcl-2-associated X protein(Bax)(P＜0.001),while Bcl-2,the Bcl-2/Bax ratio,and calcein-AM green fluorescence intensity were decreased(P＜0.001).Compared with the model group,the Chinese medicine group,nicorandil group,and valsartan group showed decreased mRNA expression levels of caspase-3,caspase-9,Cyt-c,Bad,and Bax(P＜0.05,P＜0.01,or P＜0.001),increased Bcl-2 and Bcl-2/Bax ratio(P＜0.001),enhanced calcein-AM green fluorescence intensity(P＜0.05 or P＜0.001),and increased JC-1 aggregate/monomer ratio(P＜0.05,P＜0.01,or P＜0.001).Conclusion:The Liqi Huatan Huoxue Formula can inhibit cardiomyocyte apoptosis by upregulating NRG-1 expression and mediating the mitochondrial apoptosis pathway.关键词
