二陈汤对小鼠结直肠癌皮下移植瘤模型的作用机制研究
Mechanisms of Erchen Decoction on Subcutaneous Transplanted Tumor Model of Colorectal Cancer in Mice

Abstract
Objective:To investigate the effects and underlying mechanisms of Erchen Decoction on subcutaneous MC38 colorectal cancer xenografts.Methods:Twenty-two C57BL/6 mice were randomly divided into a control group and an Erchen Decoction group.All mice were inoculated with MC38 colorectal cancer cells.The Erchen Decoction group received oral administration of Erchen Decoction for three weeks.Body weight,tumor volume,and tumor weight were recorded.Hematoxylin and eosin(HE)stai-ning was used to observe histopathological changes.Immunohistochemistry(IHC)was performed to detect the expression of F4/80 antigen(F4/80),inducible nitric oxide synthase(iNOS),cluster of differentiation 206 and 3(CD206,CD3),Ki-67 antigen(Ki67),CD4,CD8,and forkhead box protein P3(FoxP3)in tumor tissues.Flow cytometry(FCM)was used to assess the expression of F4/80,major histocompatibility complex class Ⅱ(MHC-Ⅱ),and CD206.Quantitative real-time PCR(qPCR)and Western blot(WB)were conducted to detect the mRNA and protein expression of tumor necrosis factor-alpha(TNF-α)and arginase-1(Arg1).Re-sults:Compared with the control group,mice in the Erchen Decoction group showed significantly reduced body weight,tumor vol-ume,and tumor mass(P<0.05).Protein expression of F4/80,CD206,Ki67,and FoxP3 was decreased,while iNOS expression was increased(P<0.05).The proportion of F4/80-positive cells and the mRNA and protein expression levels of Arg1 in tumor tissues were also reduced(P<0.05).There were no statistically significant differences between the two groups in the expression of CD3,CD4,CD8,MHC-Ⅱ,CD206,or TNF-α(P>0.05).Conclusion:Erchen Decoction inhibits the growth of subcutaneous MC38 tumors in mice,possibly by reducing the recruitment and M2 polarization of tumor-associated macrophages.关键词
