清心宁神汤调控磷脂酰肌醇3-激酶信号通路治疗不寐的作用机制
Mechanisms of Qingxin Ningshen Decoction in Treating Insomnia by Regulating Phosphatidylinositol 3-kinase Signaling Pathway

Abstract
Objective:To explore the potential mechanisms of Qingxin Ningshen Decoction in treating insomnia caused by heart failure in housing the spirit in rats.Methods:A model of insomnia caused by heart failure in housing the spirit was established in SD rats using caffeine combined with 4-chloro-DL-phenylalanine(PCPA).Rats were randomly divided into normal group,model group,zolpidem group,low-dose Qingxin Ningshen Decoction group,high-dose Qingxin Ningshen Decoction group,and pathway in-hibitor group,six rats in each group.The synergistic hypnotic effect of the drugs was assessed by the pentobarbital sodium sleep po-tentiation test.Protein expression levels of phosphoinositide 3-kinase(PI3K),protein kinase B(AKT),forkhead box O(FOXO),and their corresponding phosphorylated proteins in rat brain tissue were detected by Western blot.The mRNA expression levels of PI3K,AKT,and FOXO were measured by real-time quantitative PCR(qPCR).Immunohistochemistry(IHC)and immunofluores-cence(IF)assays were used to evaluate the expression and phosphorylation levels of PI3K,AKT,and FOXO in brain tissue.Re-sults:Compared with the model group,the zolpidem group,low-and high-dose Qingxin Ningshen Decoction groups,and pathway in-hibitor group showed significantly shortened sleep latency(P<0.01)and prolonged sleep duration(P<0.01).Expression levels of PI3K,AKT,FOXO,and their phosphorylated proteins and mRNA in brain tissue were decreased(P<0.05 or P<0.01).IHC and IF results demonstrated reduced expression levels and fluorescence area of PI3 K,AKT,FOXO,and corresponding phosphorylated proteins(P<0.05 or P<0.01).Conclusion:Qingxin Ningshen Decoction can improve sleep in rats by inhibiting the expression of factors in the PI3K/AKT/FOXO signaling pathway,with the high-dose group showing the best therapeutic effect.关键词
