蛭龙活血通瘀胶囊改善心肌缺血再灌注损伤的机制研究
Mechanism of Zhilong Huoxue Tongyu Capsules in Improving Myocardial Ischemia-reperfusion Injury

Abstract
Objective:To investigate the effect of Zhilong Huoxue Tongyu Capsules(abbreviated as Zhilong)on myocardial ische-mia-reperfusion injury(MIRI).Methods:Forty mice were randomly divided into sham operation group,model group,low-dose Zhi-long group,and high-dose Zhilong group using a random number table.Pretreatment with the drug was administered for two weeks before model establishment.A modified thread embolization method was used to establish the MIRI model.Serum levels of malondi-aldehyde(MDA),lactate dehydrogenase(LDH),creatine kinase-MB isoenzyme(CK-MB),cardiac troponin T(cTnT),and interleu-kin-33(IL-33)were measured by enzyme-linked immunosorbent assay(ELISA).Myocardial infarct size was observed by 2,3,5-tri-phenyltetrazolium chloride(TTC)staining.Hematoxylin-eosin(HE)staining was performed to assess pathological changes in heart tissue.Apoptosis was detected by terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)assay.Immunofluores-cence was used to observe the expression of ST2 and IL-33 in myocardium.Western blot(WB)was applied to detect protein ex-pression levels of ST2,IL-33,B-cell lymphoma 2(Bcl-2),and Bcl-2-associated X protein(Bax)in myocardial tissue.Results:Compared with the sham operation group,the model group showed significantly increased serum levels of MDA,LDH,CK-MB,cT-nT,and IL-33(P<0.01),a large white infarct area in the myocardium(P<0.01),increased apoptotic cells(P<0.01),and ele-vated myocardial expression of ST2 and IL-33(P<0.01).Protein expression levels of ST2,IL-33,and Bax in myocardial tissue were increased(P<0.01),whereas Bcl-2 expression was decreased(P<0.01).Compared with the model group,the high-dose Zhilong group showed reduced serum levels of MDA,LDH,CK-MB,cTnT,and IL-33(P<0.01),decreased infarct size(P<0.05),reduced myocardial tissue damage and apoptosis(P<0.01),decreased myocardial ST2 and IL-33 expression(P<0.01),and lower protein expression levels of ST2,IL-33,and Bax(P<0.01),while Bcl-2 expression was increased(P<0.01).Conclu-sion:Zhilong Huoxue Tongyu Capsules may improve MIRI by inhibiting inflammatory mediator expression and cell apoptosis through the IL-33/ST2 signaling pathway.关键词
