淫羊藿苷对肱骨骨折大鼠破骨细胞活性和血管形成的影响和作用机制
Effects and Mechanisms of Icariin on Osteoclast Activity and Angiogenesis in Rats with Humeral Fracture

Abstract
Objective:To investigate the effects of icariin(ICA)on osteoclast activity and angiogenesis in humeral fracture rats based on the vascular endothelial growth factor(VEGF)/extracellular signal-regulated kinase 1/2(ERK1/2)signaling pathway.Methods:Sixty male Sprague Dawley(SD)rats were randomly divided into six groups(n=12 per group),including normal group,model group,ICA low-dose group(40 mg/kg),ICA high-dose group(80 mg/kg),and ICA high-dose+VEGF inhibitor group(80 mg/kg ICA+5 mg/kg bevacizumab).A humeral fracture model was established using bone clamp osteotomy combined with Kirschner wire intramedullary fixation.After 21 days of drug intervention,fracture healing was evaluated by X-ray.Humeral bone mineral density was measured by dual-energy X-ray absorptiometry(DEXA).Serum levels of VEGF,basic fibroblast growth factor(bFGF),and angiopoietin-1(ANG-1)were measured by enzyme-linked immunosorbent assay(ELISA).Histological changes and new vessel counts in humeral tissue were assessed by hematoxylin-eosin(HE)staining.Osteoclast numbers were detected by tar-trate-resistant acid phosphatase(TRAP)staining.Protein expression levels of VEGF,ERK1/2,phosphorylated ERK1/2(p-ERK1/2),and osteogenesis-related proteins Runt-related transcription factor 2(Runx2)and alkaline phosphatase(ALP)in bone tissue were determined by Western blot.Results:Compared with the model group,both ICA low-and high-dose groups showed gradual dis-appearance of fracture lines,increased callus formation,increased humeral bone mineral density,elevated serum VEGF,ANG-1,bF-GF levels,increased number of new vessels,and higher protein expression levels of VEGF,p-ERK1/2/ERK1/2,Runx2,and ALP in bone tissue,along with decreased osteoclast numbers(P<0.05).The VEGF inhibitor bevacizumab significantly attenuated the pro-moting effects of ICA on fracture healing in rats.Conclusion:ICA promotes angiogenesis,improves osteoclast activity,and acceler-ates fracture healing in humeral fracture rats,potentially through activation of the VEGF/ERK1/2 signaling pathway.关键词
