祛风消癜方对过敏性紫癜大鼠辅助性T细胞17/调节性T细胞的影响
Effects of Qufeng Xiaodian Formula on Th17/Treg Balance in Rats with Allergic Purpura

Abstract
Objective:To investigate the effects of Qufeng Xiaodian Formula on T helper 17(Th17)and regulatory T(Treg)cells in a rat model of allergic purpura.Methods:Forty-five male Sprague Dawley(SD)rats were randomly divided using a random num-ber table into a control group(n=9)and a modeling group(n=36).After six weeks of model induction,the rats in the modeling group were further randomly divided into the model group,compound glycyrrhizin group,and low-,medium-,and high-dose Qufeng Xiaodian Formula groups(n=6 per group).The control and model groups received 0.9％sodium chloride solution by gavage.The compound glycyrrhizin group received 1.34 mg/mL compound glycyrrhizin suspension.The low-,medium-,and high-dose Qufeng Xiaodian Formula groups received 0.74 g/mL,1.48 g/mL,and 2.96 g/mL Qufeng Xiaodian Formula suspensions,respectively.All treatments were administered once daily for four weeks.At the end of week 10,the Th17/Treg ratio in rat kidney tissue was meas-ured,along with interleukin-6(IL-6)/signal transducer and activator of transcription 3(STAT3),serum interleukin-2(IL-2)/STAT5,retinoic acid-related orphan receptor gamma t(ROR-γt),and forkhead box protein P3(FOXP3)mRNA and protein expres-sion levels.Results:Compared with the model group,all treatment groups showed significantly decreased levels of renal Th17 cells,Th17/Treg ratio,STAT3,FOXP3 mRNA and protein expression,serum IL-6,transforming growth factor-beta1(TGF-β1),and IL-17 levels(in the high-dose group only),while Treg cell levels,STAT5,and FOXP3 mRNA and protein expression were significantly in-creased(P＜0.05).Conclusion:The Qufeng Xiaodian Formula can regulate the Th17/Treg imbalance in allergic purpura rats,mainly through modulation of the IL-6/STAT3 and IL-2/STAT5 signaling pathways.关键词
