清热化痰理气方及其拆方改善细颗粒物肺损伤的机制研究
Mechanism of Qingre Huatan Liqi Formula and Its Disassembled Prescriptions in Ameliorating Fine Particulate Matter-induced Pulmonary Injury

Abstract
Objective:Based on neutrophil extracellular traps(NETs),this study aimed to investigate the mechanisms by which the Qingre Huatan Liqi Formula and its disassembled formulas(Qingre Jiedu Formula and Liqi Huatan Formula)ameliorate fine partic-ulate matter(PM2.5)-induced lung injury.Methods:A lung injury rat model was established via intratracheal instillation of PM2.5 suspension.Rats were divided into six groups:control,model,Qingre Jiedu,Liqi Huatan,Qingre Huatan Liqi,and erythromycin pos-itive control.Hematoxylin-eosin(HE)staining was used to observe lung tissue pathology.ELISA and quantitative real-time PCR(qPCR)measured interleukin-1β(IL-1β)protein and mRNA expression.ELISA also detected NET components[myeloperoxidase-DNA complexes(MPO-DNA),neutrophil elastase(NE),citrullinated histone H3(CitH3)]and NET regulatory enzymes[reactive oxygen species(ROS),NADPH oxidase,peptidylarginine deiminase 4(PAD4)].Double-stranded DNA(dsDNA)content was quantified using Picogreen fluorescence assay,and Western blot analyzed NAD(P)H dehydrogenase[quinone]1(NQO1)and MAPK pathway-related proteins.Results:All medication groups showed improved lung injury,with reductions in NET components,NET regulatory enzymes,and airway inflammatory markers.The Qingre Huatan Liqi Formula exhibited superior efficacy compared to its disassembled formulas.Specifically,the Qingre Huatan Liqi group showed greater reduction in ERK signaling activity compared to the Qingre Jiedu group and a more pronounced decrease in P38 signaling activity compared to the Liqi Huatan group(P<0.05).Conclusion:Qingre Huatan Liqi Formula and its disassembled formulas alleviated PM2.5-induced lung injury by inhibiting NET formation through downregulation of the NADPH/ROS/histone citrullination axis and ERK/P38 MAPK signaling pathways,with the integrated formula demonstrating the optimal therapeutic effect.关键词
