茯苓多糖对肺腺癌A549细胞生物学行为的影响
Effects of Poria cocos Polysaccharides on Biological Behaviors of Lung Adenocarcinoma A549 Cells

Abstract
Objective:To investigate the effects of Poria cocos polysaccharides(PCP)on the biological behaviors of lung adenocar-cinoma A549 cells and the Janus kinase-1(JAK1)/signal transducer and activator of transcription 3(STAT3)signaling pathway in non-small cell lung cancer(NSCLC).Methods:The effect of PCP on A549 cell viability was assessed using the methyl thiazolyl tetrazolium(MTT)assay to determine subsequent experimental concentrations.Cell proliferation,migration,invasion,and apoptosis were evaluated using colony formation,scratch,Transwell assays,and flow cytometry,respectively.The mRNA levels of proliferation-related genes were measured by PCR,and protein expression levels associated with migration,invasion,apoptosis,and the JAK1/STAT3 pathway were detected by Western blot.Results:Treatment with PCP at 30,40,and 50 mg/mL significantly reduced A549 colony formation rate,scratch closure rate,number of invading cells,and expression levels of cyclin D1 mRNA,c-Myc mRNA,N-cadherin,vimentin,matrix metalloproteinase-2(MMP-2),MMP-9,Bcl-2,phosphorylated JAK1(p-JAK1),and phosphorylated STAT3(p-STAT3)compared with the control(0 mg/mL PCP)(P<0.05).In contrast,apoptosis rate and protein levels of E-cad-herin,cleaved caspase-3,and Bax were significantly increased(P<0.05),exhibiting dose-dependent effects(P<0.05).Conclu-sion:PCP inhibits the malignant biological behaviors of A549 cells by downregulating JAK1/STAT3 signaling activity and modula-ting cell proliferation,migration,and invasion.关键词
