电针对胰岛素抵抗肥胖大鼠的治疗作用机制
Effects of Electroacupuncture on Treating Obese Rats with Insulin Resistance

Abstract
Objective:To observe the effect of electroacupuncture on treating obese rats with insulin resistance(IR).Methods:Wistar male rats were randomly divided into the normal group,model group,electroacupuncture group,and sham electroacupuncture group,with eight rats in each group.Except for the normal group,other groups were fed with high-fat feedstuff to build an IR obese rat model.The rats in the electroacupuncture group were given electroacupuncture intervention,while the rats in the sham electroa-cupuncture group were given sham electroacupuncture invention,with the same grasping and fixation only given to the normal and model groups.The intervention was conducted once every other day and three times a week for eight consecutive weeks.Before and after intervention,the body weight,and anal and nasal length of rats were measured,with Lee's index calculated.After the interven-tion,the weight of perirenal fat and epididymal fat was measured.Meanwhile,intraperitoneal glucose tolerance test(IPGTT)was performed to measure blood glucose,and the glucose infusion rate(GIR)was calculated by adopting the hyperinsulinemic glucose clamp technique.Flow cytometry was utilized to detect the percentage of Th17 and Treg cells in the adipose tissue and peripheral blood,and calculate Th17/Treg.Western blotting was leveraged to detect the expression of interleukin-17 receptor(IL-17A)and FOXP3 proteins in the adipose tissue of rats.Results:Compared with the model group,the body weight,Lee's index,perirenal fat weight,and epididymal fat weight in the electroacupuncture group were low,with low IPGTT blood glucose and high GIR.Addition-ally,the percentage of Th17 cells in adipose tissue and peripheral blood was low,while that of Treg cells was high and the Th17/Treg value was low,with low expression of IL-17A protein in adipose tissue and high expression of FOXP3 protein(P<0.05).Conclusion:Electroacupuncture can reduce the body weight,perirenal fat,and epididymal fat weight of obese rats,and improve in-sulin sensitivity,which may be related to the regulation of Th17/Treg cell balance by electroacupuncture.关键词
