浙贝母-土贝母药对对乳腺癌的抑制作用及机制研究

Abstract
Objective:To investigate the inhibitory effect and mechanism of the Fritillariae Thunbergii Bulbus-Bolbostemmatis Rhi-zoma drug pair on tumors in human breast cancer 4T1 cell nude mice.Methods:A human breast cancer 4T1 cell tumor-bearing nude mouse model was established and randomly divided into a model group,a positive control group,and high-,medium-,and low-dose Fritillariae Thunbergii Bulbus-Bolbostemmatis Rhizoma groups(5 mice per group).The high-,medium-,and low-dose groups received raw drug solution at 5.2 g/mL,2.6 g/mL,and 1.3 g/mL,respectively,by gavage,at 0.1 mL/10 g once daily.The model group received an equal volume of physiological saline once daily,and the positive control group received 0.1 mg of adriamycin via intraperitoneal injection once every 7 days.All treatments continued for 21 days.At 24 hours after the last administration,mice were anesthetized with 1%pentobarbital sodium,and tumor tissues were harvested,photographed ex vivo,and measured to calculate tumor mass and tumor inhibition rate.Protein expression of Janus kinase(JAK)1,JAK2,and signal transducer and activator of transcription 3(STAT3)in tumor tissues was detected by Western blot.Results:Compared with the model group,all dosing groups significantly inhibited tumor growth(P<0.05).Tumor inhibition rates in the low-,medium-,and high-dose groups were 8.83%,28.63%,and 50.05%,respectively,with statistically significant differences(P<0.05).Protein expression of JAK1,JAK2,and STAT3 was significantly reduced in all dosing groups compared with the model group(P<0.05).Conclusion:The Fritillariae Thunbergii Bulbus-Bolbostemmatis Rhizoma drug pair inhibits the growth of human breast cancer 4T1 cell tumors in nude mice,and its mechanism may involve modulation of the JAK/STAT pathway to inhibit tumor stem cells.关键词
